NOTE: The new guide thread can be found HERE!

So, here's the new and improved "Semi-comprehensive SMBO Guide", current as of the most recent update.  Please fill me in on missing or incorrect data whenever possible.

To save you undue scrolling, here are links to a few of the highlights of the guide:
World Map - Self-explanatory. (Thanks to Garro for most of the latter half!)
E.N.E.M.I.E.S. - Enemy stats and KO-ing values.
B.A.D.J.E. - Badges' stats and relative comparisons.
R.E.C.I.P.E. - Recipes' components, stats, and profitablity compared.
[New!] A.P.P.A.R.E.L. - Characters' base and equipment stats compared.

Characters

Note that "Total Stat Ratings" are calculated from initial stats and the best character-specific equipment to date. For a breakdown of the stats of these items, as well as an explanation of Stat Ratings, look at the Equipment section below.

Mario’s built for SPEED, and an average character in many respects. Mario has few defined strengths or weaknesses, and is probably a good choice for beginners.

Pros:
- Highest naturally acquired FP, DEF (tied), SPEED and SP
- Highest all-around stat total
- One of two characters with a ranged special that costs only 2 FP
- Tied for best DEF-boosting special

Cons:
- Tied for lowest naturally acquired HP
- Worst healing special
- Lack of diversity in special attacks

Specials:
Fireball (5 Damage, 3 Range, 2 FP)
Super Fireball (9 Damage, 3 Range, 4 FP)
Defense Boost (1.4x Multiplier, 1 Range, 4 FP)
Refresh (5 HP restored, 1 Range, 7 FP)
Ultra Fireball (15 Damage, 3 Range, 9 FP)

Total Stat Rating: 32.0

Though he shares several specials and some equipment with his older brother, Luigi’s one-to-one HP-for-FP trade sets him apart from the crowd.  In addition, Luigi has reasonable equip requirements, cheap equipment, and an affinity for DEF and STACHE.  With no major weaknesses, Luigi is one of the best characters in the game.

Pros:
- Second-highest naturally acquired HP and tied for highest DEF
- Discounting SP, second-highest all-around stat total (first with a non-ranged weapon)
- Second-best healing special, in convenient 5-for-5 interval
- Tied for best DEF-boosting special

Cons:
- Generally FP-ineffective special attacks
- Expensive equipment, coupled with tied-for-lowest total STACHE

Specials:
Mushroom Heal (5 HP restored, 1 Range, 5 FP)
Fireball (7 Damage, 3 Range, 4 FP)
Jump (10 Damage, 3 Range, 7 FP)
Defense Lift (1.4x Multiplier, 1 Range, 4 FP)
Defense Drain (0.7x Multiplier, 3 Range, 8 FP)

Total Stat Rating: 29.6

The buff and burly Wario’s main objective is to smash his opponents into the ground.  In exchange for his naturally high ATK, however, Wario is slow, and compared to the rest of the cast, quite defenseless.  Wario's lack of affinity for DEF and late-earned heal special make him awkward to play as at first, but in the long run, he's a fairly average character.  His ranged weapon can be a money hog, though, so you might opt to keep the one below it for when you're running short on cash.

Pros:
- By far the highest naturally acquired ATK
- Longest-ranged weapon, with a range of 3
- Cheap equipment
- Most powerful healing special
- Second-best ATK-boosting special
- Discounting SP, highest all-around stat total

Cons:
- Lowest naturally acquired DEF, SPEED (tied), and SP
- Four of five specials have only 1 Range

Specials:
Wario Ram (4 Damage, 1 Range, 2 FP)
Muscle Growth (1.3x Multiplier, 1 Range, 4 FP)
Bob-Omb Toss (10 Damage, 3 Range, 6 FP)
Defense Down (0.8x Multiplier, 3 Range, 5 FP)
Carrot Candy (9 HP restored, 1 Range, 10 FP)

Total Stat Rating: 29.4 (25.9 w/o ranged weapon)

Much like in mainstream Mario games, Waluigi’s a rather awkward character to deal with.  His main focus is on STACHE, giving him a lucrative glass-cannon playstyle. His specials, save for the first, are rather nondescript, but otherwise, he has no obvious disadvantages.  In addition, his ranged weapon can be quite useful in dispatching of enemies from a distance.

Pros:
- Highest naturally acquired STACHE and second-highest SPEED
- Cheapest ATK-boosting special
- Cheapest 4-Range special
- One of two characters with a ranged weapon, and has the cheapest ammo
- Has a third weapon comparable to his fourth, which can be used to boost total base+equip DEF to 7

Cons:
- Naturally acquired FP of 0
- Second-lowest all-around stat total
- Second-worst healing special

Specials:
Waluigi Rush (1.2x Multiplier, 1 Range, 2 FP)
Sinister Slash (8 Damage, 4 Range, 5 FP)
Sly Rejuvenation (6 HP restored, 1 Range, 7 FP)
Attack Down (0.8x Multiplier, 3 Range, 4 FP)
Diabolical Slash (14 Damage, 4 Range, 8 FP)

Total Stat Rating: 27.0

Yoshi is easily the best character to play as, as his affinity for FP and phenomenal special attacks often work together to greatly simplify otherwise difficult encounters.  Yoshi has the only heal special that gives a greater than 1-to-1 exchange of HP for FP, making leveling on Lakitus and such quite lucrative.  Yoshi has no defined disadvantages, and as an added bonus, you can use spray paint to further customize your character.

Pros:
- Tied for highest naturally acquired DEF, second-highest ATK and FP
- One of two characters with a ranged special that costs only 2 FP
- Best healing special (7-for-6)
- Best ATK-boosting special
- Highest damage/cost ratio in specials

Cons:
- Extremely expensive equipment
- Tied for lowest naturally acquired HP

Specials:
Egg Pelt (6 Damage, 3 Range, 2 FP)
Egg Restore (7 HP restored, 1 Range, 6 FP)
Egg Charge (1.4x Multiplier, 1 Range, 5 FP)
Weak Yoshi Bomb (15 Damage, 4 Range, 7 FP)
Super Egg Pelt (18 Damage, 4 Range, 8 FP)

Total Stat Rating: 28.1

Toad’s an unlikely fortress, as his naturally high HP and DEF make for an interesting time for your opponents.  Coupling that with his fourth special, Toad easily has the most of control over PvP.  Toad’s most prominent weakness is his general lack of ATK, but that can be remedied through the aforementioned special.  Toad’s hard to place in the lineup for character ranking, as his performance is largely based on your desired strategy.

Pros:
- Highest naturally acquired HP
- Has the unique ability to swap ATK & DEF
- Cheapest DEF-boosting special

Cons:
- Lowest naturally acquired ATK, tied for second-lowest FP
- Only character with no healing special
- Generally nondescript and cost-ineffective specials
- By far the lowest all-around stat total

Specials:
Mushroom Barrier (1.2x Multiplier, 1 Range, 2 FP)
Mushroom Toss (6 Damage, 3 Range, 5 FP)
Super Shroom Toss (11 Damage, 3 Range, 7 FP)
Attack-Defense Swap (1 Range, 4 FP)
Ultra Shroom Toss (14 Damage, 3 Range, 8 FP)

Total Stat Rating: 25.7

Character Tier (rough and subject to change):
Yoshi
Luigi

Wario
Mario

Waluigi
Toad

For a brief comparison of the characters' stats, as well as their equipment's, check out this brand-new, snazzy image, called "All Player-specific Paraphernalia Analyzed in Relative Efficiency by Level", or "A.P.P.A.R.E.L." for short:

Linked for size

The Stat Rating system is broken down in the Equipment section below, and the "Total" chart includes a character's base stats and best (latest earned) equipment.  SP gained from level-ups and SPEED was not factored into the total SP values shown.

Here’s a chart of the “Relative Effectiveness of Cash Influx from the Preparation of Eats” (R.E.C.I.P.E.), detailing how much of a profit you can get by making each recipe, as well as concisely listing all of the recipes and their stats.  Note that prices are always in the format (0 STACHE / 30 STACHE), and generally scale approximately linearly.

Linked for size

All buy prices are the cheapest possible if applicable, or the item’s sell price if it can’t be bought.  Note that the Melon’s price is based on its relative value to Drillbit Shells from the KIV favor, and the Cake Mix’s price is determined by its sell price; the actual worth of these items are 1 DE coin and 1 Dim Star Piece, respectively.

Equipment

Equipment in SMBO is currently divided in to seven subcategories: Armor, Leg Armor, Weapons, Headwear, and Mushroom, Flower, and Special Badges.  "Stat Ratings", placed below each entry, are an approximate measure of the worth of equipment.  Each 1.0 Stat Rating = 5 HP, 5 FP, 1 ATK, 1 DEF, 2 SPD, 2 STA, 5 SP, or 3% Critical or Block Chance.

World Map & Favors

This is a semi-comprehensive map of the game as of its most recent update. For better quality, follow the direct link here.

For the first favor, give the Toad at the point indicated three Koopa Shells to gain access to the warp in the dark purple house, and thusly the HP Plus Badge.
For the second, talk to the Toad in the indicated location, then kill 10 Lakitus in the same screen (not necessarily in one life).  Talk to the Toad again to receive 100 coins.
For the third, talk to the Toad, then head to the screen with seven Piranha Plants and look behind a tree for the Mushroom Toy.  Give this to the Toad to gain access to warp #7, and the second half of Koopa Beach.
For the fourth, talk to the Salestoad to find out that his sick friend has his goods in a package.  Go to the Restoration House and talk to the Toad in the right corridor to have him give you the package.  Return it to the first Toad to gain access to the shop containing the third set of equipment.
For the fifth, talk to the Koopa in the house indicated and give him three Drillbit Shells in exchange for five Melons, rare items which stack and give 10 HP apiece.
For the sixth, retreive the envelope for Toadsdale after hitting the four switches.
For the seventh favor, deliver the Toad ten Pokey Flowers, in exchange for 200 experience points.
For the eighth, talk to the Toad in the northwest corner of the town, then talk to the other Toads to eventually receive the Goomba Toy. Give this to the first Toad to receive a Pokey Card.
For the ninth favor, deliver the green-capped Toad the package of Super Shroom (located in the upper-right corner of the screen two screens west of the vine pit.
For the tenth, deliver a Shroom Cake to the Toad outside of Flower Hills.  In exchange, he'll allow you to sample his wares.

Other notes:
- The three points marked in the Mushroom Maze inset are a Moderately Weak Mini Bob-Omb, a Moderate Mini Bob-Omb, and the point where the former is used, respectively.
- Warp 16 is a Bullet Bill cannon, and is illustrated as a yellow/red warp for simplicity’s sake.  To make the Golden Bullet Bill appear, you will first need to find the clues to its location, the first being located around cacti in the bottom-right of the screen one screen east of the cannon, and the second in front of a small cliff in the screen one screen south of the cannon.
- The spray can opposite the card shop is where Yoshi and Toad can have their skin/cap color changed.

Here is a map of the very expansive Mushroom Kingdom area, with a comprehensive listing of various points of interest.

Green – Favor Locations
Blue – Sites of Interest
Red – Spin Badge Panel Locations
Purple – Mushroom Ball Locations
Grey  – Other Points of Interest

Favor Descriptions
#1 – Talk to Peach to have her enlist your help in finding her precious item.  Return it to her to receive 2,000 coins, a Deluxe Meal, and a very helpful badge.
#2 – Breach the lines of defense this Koopa has set up to have him ask for the number of buildings in the Mushroom Kingdom.  Answer correctly to receive a rare badge.
#3 – Bring this Goomba a Jelly Super to receive 200 Coins and a Star Piece.
#4 – Return this Toad’s Mushroom Balls to receive a Blue Candy and 3 Star Pieces.

Sites of Interest
#1 – Restoration House.
#2 – Item Shop & Item Holder.
#3 – Badge Bazaar & Star Piece Exchange.
#4 – Equipment Shop.
#5 – Dodgebill Arcade.
#6 – A locked house used in one of the favors.
#7 – A locked house containing one of the favors.
#8 – The Cake Mix vendor’s house.
#9 – Salt T.’s kitchen.
#10 – Peach’s Castle.

Other
#1-#3 - ? Blocks.  The second one is the most elusive, and requires the use of a hidden Drill Badge panel to reach.
#4 – Shortcut to the beginning of the MK path.  Requires a Hammer Barrage badge.
#5 – The Small Key is located in this area, and is required for one of the favors.
#6 – Save Album.
??? – This house is linked to a very secret item. Bringing it here will grant you a unique reward.

And last, but not least, here's a map of the right path of the Daily Event, commonly referred to as the "DE Maze" or simply the "hard side".

Legend should explain it all; there are three DE Koopas before the pipe, and five afterward.

Again, please let me know if there are any glaring errors in this guide.  Screenshot proof (try ScreenHunter Free 5.0) is always most helpful to illustrate a complaint.

Awesome guide, it looks great!

Just something I want to point out, the color-changer and golden bullet bill icons aren't on the Legend...

Additionally, to prevent eight-then-parentheses from looking like this: Cool

Click Modify Post, then go to Additional Options. Check off 'Don't use smileys.' and it won't show up as a smiley.

I realize that certain things aren't on the legend, but I figured they were self-explanatory.  I might change them to "Points of Interest" or such, and mark them with a differently-colored number (white, mayhaps?).

EDIT: Thanks, BlazeingYoshi, for the recipe info.  I'll get on that right away.

Wow! I really like your character descriptions! However, I wouldn't underrate Wario so much; we haven't had enough Wario players to come up with a good strategy yet.

Overall, terrific job!

Well, a somewhat-good strategy for a Wario is maybe to provide backup for other characters- he can't really be a tank, due to low DEF and STA bonuses, so he could be a support character- buff a character with his special whilst the character uses his, and then use Defense Down on the enemy, run away, then attack with the Bob-omb Shooter.

Other than debuffing foes (might work quite well with Waluigi's ATK Down [but then people'd just run, unless it was turn based PvP*]), he can't really do much.

|contemplates deleting his Wario and making a Toad... again|

*If it's ever made.

It just seems that there isn't much that Wario can do better than other characters, and he has a very low affinity for DEF, meaning that more level-ups will have to be poured into it.

Yoshi and Luigi, on the other hand, have strengths in good stats other than ATK, and thus their stat allocations are far more malleable.  The only difference between the two of them is that Yoshi's specials are a bit better (predominantly Yoshi's ATK-booster versus Luigi's DEF-booster); everything else is actually rather comparable.

Toad I haven't played as, and I can't seem to figure out exactly how he'd work if played.  I imagine he's got a lot of control over PvP, what with his massive HP and DEF and his ATK-DEF switching special.

EDIT: Finally deleted the old guide, derp.

What level do you get the specials for each character?
Specifically Toad's Mushroom Toss and Yoshi's Egg Pelt. Huh?

The first special is earned at level 5 or 6, and subsequent specials are usually about four levels after the preceding one.

Also, revamped the Recipes/Items section slightly, including a newly discovered recipe courtesy of Shroomerson.

Amanda and I figured out what the Gnarantula Cola recipe was:

Blue Gnarantula Cola (Doubles Defense for 10 seconds) (-, 40(I assume this is the base price to sell)): Red Gnarantula Cola + Tough Koopa Shell.

EDIT: Amanda found it first, I just went to confirm it.

Thanks a bunch, I'll add it and the other two confirmed ones to the guide when I update the enemy stat section.  I'll verify the sell price, as well.

No need to give me the sell price for Shroom Cake and Cake, BTW.
